MPY NUPharose HP and NuPerley MPY are sulfur affinity fillers formed by bonding sulfur-containing compound 2-mercaptopyridine (MPY for short) on agarose gel microspheres. MPY groups contain sulfur atoms and pyridine groups, which can form sulfur-affinity and hydrophobic interactions with target molecules, thus being classified as multimodal fillers. The principle of sulfur affinity is to use the interaction between electron donor and electron acceptor to separate and purify biological molecules. This force is strengthened in high salt environment and weakened in low salt environment, which is similar to hydrophobic interaction. NUPharose HP are highly cross-linked small particle size (~ 35 μm), high resolution agarose microspheres, NuPerley a new generation of highly rigid agarose microspheres (~ 60 μm). The ligand density of the two fillers is also different. The ligand density of MPY NUPharose HP is about 3.5 mg/mL, and the ligand density of NuPerley MPY is about 2 mg/mL, which provides different affinity choices. Relatively speaking, MPY NUPharose HP is mostly used in the purification of supercoiled plasmid DNA, NuPerley MPY is used in the purification of IgM and IgY. In many cases, the two fillers can achieve the same purification goal.
