Strep-Tactin 2 NUPharose FF is a mutant of streptavidin (Strep-Tactin 2 ligand) and NUPTEC bio-NUPharose sepharose coupled to form and chromatographic separation medium for isolation and purification of Strep II or Twin Strep tag proteins. The Strep II tag is a small tag (WSHPQFEK) of 8 amino acids, which generally does not affect the structure and function of the fusion protein, and is commonly used in the detection and purification of fusion expression proteins. Twin Strep tag is a tag of two Strep II in tandem. Compared with Strep II tag, Twin Strep tag has higher affinity with Strep-Tactin 2 ligand. The Strep-Tactin 2 ligand of this product has a further increased affinity for Strep II tags and a significantly reduced affinity for biotin compared to the previous generation of Strep-Tactin ligands. Therefore, this product can bind the Strep II fusion protein more firmly, and when the fusion protein expression is low, the relative capture amount is higher, and the load and yield are higher. In terms of elution, biotin can be used for elution, which is more economical than the expensive desthiobiotin elution, and can be regenerated with 10-50 mM NaOH after use. This product is highly specific for the Strep II tag and generally requires only one step of purification to obtain highly pure protein samples.
