IMAC NUPharose FF
Ni-NTA NUPharose FF and IMAC NUPharose FF are metal chelating fillers containing nitrilotriacetic acid (NTA) groups, which are mainly used for Affinity chromatograph of histidine-tagged (His-tag) proteins. These two products are based on NUPTEC biological classic NUPharose agarose gel microspheres: IMAC NUPharose FF unchelated metal ions, and users can chelate Ni2 +, Co2 +, Cu2 +, Zn2 + and other metal ions after use; Ni-NTA NUPharose FF has chelated Ni2 +, users can directly use it.
Through the optimization of the ligand modification process, the two products have achieved a balance of affinity and specificity: the binding amount of histidine-tagged protein is more than 40 mg/mL, and the purity of the sample can reach more than 90% after one-step purification. In addition to histidine, metal chelating fillers can also form coordination bonds with tryptophan and cysteine residues, so these two fillers can also be used to purify proteins without tags but containing these amino acid residues, such as human apoferritin.
