Ni-TED NUPharose FF is a metal chelating affinity filler with the lowest drop-off rate of metal ions. The ligand is tricarboxymethyl ethylenediamine (tris-carboxymethyl ethylene diamine,TED) group, the coordination metal ion is Ni2, the NUPharose is a classic agarose gel basic microsphere developed by NUPTEC biology, and FF is the abbreviation of Fast Flow. The TED ligand of this product has good stability in the five-coordination structure formed by Ni2, and has excellent tolerance to chelating agents, reducing agents, acids, alkalis and other reagents. It can be used normally under the conditions of 10 mM EDTA, 100 mM β-ME or 50 mM DTT, and can maintain excellent chromatographic performance after being treated for 24 h under the condition of pH = 1 or pH = 14, also for long-term storage in 10 mM NaOH, the concentration of imidazole required in the eluent is lower than that of other metal chelating fillers.

Chemical stability

1) Purification buffer: commonly used aqueous buffer, which may contain 10 mM EDTA, 100 mM β-ME or 50 mM DTT.

2) Cleaning in place (CIP) treatment: 1 M NaOH, 1~2 M NaCl, 70% ethanol, 30% isopropanol, 0.1~0.5% non-ionic surfactant. 3) The performance of the resin remains stable after treatment in the following solutions for a specific time: 10 mM NaOH (long-term), 0.1 M hydrochloric acid (24 h), 6 M guanidine hydrochloride (24 h).
