Easily eluted protein G

Protein G is a protein in the cell wall of streptococci that specifically binds to the Fc segment of the immunoglobulin IgG molecule. It not only binds to all subclasses of human, mouse and rat IgG, but also to IgG of guinea pig, rabbit, goat, cow, sheep and horse. The use of protein G is extremely wide, can be used to separate and purify or remove IgG from serum, ascites, purification of monoclonal antibodies and separation of antigen-antibody complexes. Natural protein G is composed of multiple similar domains. Under neutral conditions, it has high affinity to Fc fragment of IgG. As pH decreases, the affinity gradually decreases. However, it still needs a very low pH to separate IgG from protein G. For example, it needs pH 2.5-3.5 to elute IgG from protein G medium, which is easy to aggregate and inactivate some antibodies. Easy to elute protein G(ee-ProG) optimizes the domain of natural protein G, maintains the affinity under neutral conditions, greatly reduces the affinity of acidic environment, and makes the antibody easier to dissociate and elute. Generally, the antibody can be dissociated at pH3.5-5.0.
