Streptavidin (Streptavidin,SA) and biotin have high affinity, affinity constant Kd can generally reach 10-15M, active SA usually exists in the form of tetramer. Because SA does not contain sugar, SA has a lower non-specific binding level than pro-hormone in detection applications. It has been widely used in enzyme-linked immunosorbent assay, immunohistochemistry, time-resolved immunofluorescence (TRFIA), quantitative PCR, single-stranded DNA preparation, biological molecule purification, monoclonal antibody preparation and other biotechnology fields. However, due to the ultra-high affinity of SA and biotin, it is generally difficult to separate the combined SA and biotin by mild methods. The conditions under which SA can be separated will generally make SA inactive. Similarly, biotin-labeled antibodies and antigens will also lose their activity. Therefore, it is generally believed that the binding of SA and biotin is irreversible. Recombinant streptavidin 3(r-SA3) is modified at the molecular level on the basis of natural streptavidin to greatly weaken the affinity with biotin (Kd ≈ 10-8M), while maintaining the tetramer structure, which can realize reversible binding and dissociation with biotin, and can be used for mild separation, purification and recycling of biotinylated proteins, antibodies, nucleic acids and other substances. r-SA3 containing cysteine in the molecule, directional coupling can be achieved under certain conditions.
