Enterokinase (EC 3.4.21.9) is a serine proteolytic enzyme, which can efficiently and specifically identify the sequence of Asp-Asp-Asp-Asp-Lys in protein (DDDDK), hydrolyze peptide bonds at the C- terminal of lysine (Lys,K), produce cleavage, and hydrolyze trypsinogen into trypsin in vivo. Enterokinase is widely used in the development of genetic engineering products due to its high specificity and efficient enzyme cleavage characteristics. Natural enterokinase is composed of a 115kDa heavy chain and a 35kDa light chain, the heavy chain plays the role of anchoring the cell membrane, and the light chain has the complete catalytic activity of the whole enzyme. This product is a fusion protein of the core region of the light chain activity of bovine enterokinase and histidine tag, expressed by Escherichia coli secretion technology, and has higher specific activity than natural enterokinase. At the same time with a histidine tag, after the enzyme can be used nickel column (NRPB07) to remove the rbEK-His in the sample, especially suitable for genetic engineering fusion protein enzyme digestion.

Note:

(1) Some reagents will affect the activity of the enzyme, such as> 2M Urea,>250mM NaCl,>20mM β-ME,>0.1% SDS,>50mM Imidazole. If the sample contains these components, it needs to be dialyzed to 50mM Tris-HCl (pH 8.0).
(2) Phosphate can inhibit the activity of enterokinase, and phosphate buffer should be avoided as an enzyme digestion system.
