Streptavidin (Streptavidin,SA) is a homotetrameric protein secreted by Streptomyces avidin (Streptomyces avidinii) during growth. Like avidin, one mole of streptavidin can bind four moles of biotin, with a high affinity for biotin. Since streptavidin does not contain sugar groups and the isoelectric point is close to neutral, streptavidin has a lower level of non-specific binding in detection applications than pro-biotin. Compared with natural streptavidin, the recombinant long-arm thiol core streptavidin removes the sequence irrelevant to the activity and only retains the core sequence relevant to the activity. It is superior to natural streptavidin in terms of stability and solubility. At the same time, compared with NUPTEC biological streptavidin series A09, six additional flexible amino acids are inserted before the cysteine at the C- terminal, thus lengthening the length of the thiol arm at the C- terminal, the SA can be better oriented to covalently bond with the carrier.
