The Strep II tag is a small tag (WSHPQFEK) of 8 amino acids. Because the tag is small, only about 1kDa, it generally does not affect the structure and function of the fusion protein, and is commonly used in the detection and purification of fusion expression proteins. Compared with Strep-Tactin, Strep-Tactin 2 further improves the affinity of the Strep II tag, can more firmly bind to the Strep II fusion protein, and has more advantages when used to capture the low abundance fusion protein after coupling with the immobilized material. Strep-Tactin 2 improves the affinity of Strep II, but also greatly weakens the affinity with biotin. Therefore, biotin can be used for competitive dissociation after Strep-Tactin 2 binds to Strep II fusion protein, and the elution of Strep II fusion protein can be carried out by increasing the concentration of biotin after immobilization with solid phase materials.
